Every lot is released with a Certificate of Analysis showing identity (mass spec), purity (HPLC trace) and endotoxin quantification. The CoA is filed against the lot number printed on the vial.

Peptides are synthesised in-house by Fmoc-based solid-phase peptide synthesis (SPPS) on Wang or Rink-amide resins, depending on the C-terminus. Coupling is monitored by Kaiser test; deletion sequences are minimised by double-coupling difficult residues and capping unreacted amines.

Cleavage uses a TFA cocktail with appropriate scavengers, followed by ether precipitation. Crude peptide is purified by preparative reverse-phase HPLC on a C18 column; the main fraction is collected, pooled, lyophilised and where required salt-exchanged to the acetate form to remove residual TFA.

Release testing on every lot covers:

• Identity — confirmed by ESI-MS or MALDI-TOF against the theoretical monoisotopic mass.
• Purity — measured by analytical HPLC with UV detection at 214 nm; released at ≥ 99 % HPLC purity with the chromatogram archived to the lot record.
• Appearance — visual inspection of the lyophilised cake and reconstitution behaviour.
• Endotoxin — limulus amebocyte lysate (LAL) assay where the application requires it.

Each lot ships with the Certificate of Analysis tying these numbers to the specific lot number printed on the vial. We retain a sealed retention sample for the full shelf life of the lot so any post-release question can be re-tested against the exact material that shipped.
